Inhibition of Apoptosis in Rat Colon Cancer Induced by 1, 2-Dimethylhydrazine Dihydrochloride

Abstract
The role and involvement of apoptosis were investigated in rat colon cancer model induced by 1,2-dimethylhydrazine dihydrochloride using immunohistochemical analysis of bcl-2, bax, p53 expression, and short term ex vivo culture techniques. The apoptotic loss progressively increased upto 24h after cell culture in the normal colon. The percentage of the apoptosis in the normal colon is higher than that in colon cancer. Therefore, the inhibition of apoptosis was correlated with malignant transformation of colonic epithelial cells. The bcl-2 expression significantly increased in adenomatous colon tissue compared to that in the normal colon tissue and the degree of bcl-2 expression gradually decreased as the tumor progresses to adenocarcinoma. In contrast, apoptosis-inducing protein called baa was mildly to moderately expressed in benign tissues, but not evidently expressed in adenocarcinoma. The results of this study suggested that the inhibition of apoptosis occurring in rat colon carcinogenesis may be the driving force of colon carcinogenesis.
